Principaux Thèmes de recherche : Chimie médicinale pour le traitement des cancers et des maladies neurodégénératives L’objectif de notre équipe consiste à concevoir et synthétiser de petites molécules à visée thérapeutique pour le traitement des maladies neurodégénératives et du cancer. Il ne s’agit pas tant de développer des molécules actives mais des candidats médicaments et des outils ou sondes chimiques permettant d’aller explorer des mécanismes biologiques. Ces composés sont conçus rationnellement en utilisant la modélisation moléculaire et l’expertise en chimie médicinale de l’équipe. Forte d’une longue expertise en chimie organique et thérapeutique, notre équipe travaille en collaboration étroite avec des physiciens, pharmacologues, biologistes et cliniciens. Nos efforts ont permis de découvrir un médicament, aujourd’hui sur le marché (Agomelatine, Valdoxan®, antidépresseur ligand des récepteurs mélatoninergiques commercialisé par les laboratoires Servier) et un candidat médicament actuellement en fin de phase I des essais cliniques pour le traitement de la maladie d’Alzheimer (AZP2006). Nos principaux projets actuels se focalisent sur :
1) l’étude et la validation d’une cible biologique novatrice impliquée dans la dégradation des protéines altérées pour le traitement des maladies neurodégénératives via la conception de sondes chimiques pour l’étude du mécanisme et de ligands permettant d’interagir avec cette cible,
2) le développement d’antagonistes du récepteur à l’adénosine A2a impliqué dans les maladies de Parkinson et d’Alzheimer
3) l’étude de plusieurs interactions protéine-protéine, importantes dans le traitement des cancers et la conception de petites molécules organiques susceptibles d’empêcher la formation de ces complexes protéiques.

Le CERMN occupe depuis janvier 2009 des locaux récents et fonctionnels (1420 m2) sur le campus du Pôle de Formation et de Recherche en Santé de l’Université de Caen, à proximité immédiate du CHU, du CLCC Baclesse, du Centre CYCERON et des unités de recherche qui constituent ses partenaires privilégiés (BioTICLA et GMPc). L’unité comprend 29 personnels permanents : 10 Professeurs, 5 Maîtres de Conférences (dont 2 sont HDR), 5 Ingénieurs de Recherche, 1 Ingénieur d’Etudes, 1 Assistante‐Ingénieur, 2 Techniciennes, 3 Adjoints Techniques. 4 étudiants post‐doctorants, 3 ATER et 9 doctorants viennent compléter les effectifs au 30 juin 2015.
Le CERMN constitue aujourd’hui une unité de Drug Design interdisciplinaire, à l’interface des sciences physico‐chimiques et biologiques, capable de concevoir et de synthétiser des agents d’intérêt thérapeutique ou diagnostique, dont l’activité in vitro et la drugabilité sont avérées, pour leur éventuel développement préclinique en collaboration avec des biologistes et des pharmacologues. L’unité est organisée en quatre plateformes au service de deux adressages scientifiques prioritaires dans le domaine de la cancérologie et des neurosciences. Au‐delà chaque plateforme met aussi ses outils et savoir‐faire au service de nombreuses collaborations extérieures.

Polypharmacologie
Quatreplateformesoutils L’activité de la plateforme chimie organique et médicinale est dévolue à la synthèse des molécules d’intérêt thérapeutique ou diagnostique objets des programmes de recherche. Pour cela la plateforme met au point des méthodologies innovantes de synthèse organique, notamment par la mise en œuvre de réactions de couplages métallo‐catalysées et de réactions de C‐H activation. La plateforme chémoinformatique constitue notamment un outil d’aide à la conception rationalisée de principes actifs par la modélisation moléculaire des interactions ligands‐cibles. Elle permet à travers des techniques de structure‐based ou ligand‐baseddrug design d’exploiter des chimiothèques et notamment celle de l’unité. Elle développe en parallèle une activité de data mining importante. La plateforme chimiothèque gère la collection de dérivés originaux (> 17000) issus des travaux du CERMN. Elle est annotée des résultats biologiques obtenus et constitue une banque de données inestimable utilisée en interne pour la recherche, par la fouille in silico ou in vitro, de nouveaux hits points de départ des programmes de recherche de l’unité. Incluse dans la Chimiothèque Nationale dont elle est le premier contributeur et par ailleurs devenue depuis 2013 plateau technique de la SFR ICORE de l’Université de Caen, la chimiothèque du CERMN est ouverte sur l’extérieur et développe dans ce cadre, grâce à son atelier de hit‐to‐lead chemistry, plusieurs programmes collaboratifs. La plateforme screening et drugabilité développe des tests in vitro réalisés sur un certain nombre de cibles biologiques d’intérêt (R‐5HT, cholinestérase, bêta‐A, ROS, etc…)et évalue les caractéristiques physicochimiques (solubilité, pKa, logP, passage de barrières digestive et hématoencéphalique…) prédictifs de la drugabilité des molécules issues des programmes de l’unité et propose le cas échéant des démarches chimiques ou galéniques d’amélioration de celle‐ci. Son atelier de vectorisation étudie notamment la conception de nanoparticules lipidiques ou de nanoémulsions.

LapolypharmacologieL’unité a démontré sa capacité à fédérer ses différents savoir‐faire pour le design rationnel de Multi‐TargetDirected Ligands dans le cadre de son programme PLEIAD ayant notamment permis d’identifier le donécopride comme premier agent pléiotrope, à la fois inhibiteur catalytique et périphérique de l’acétylcholinestérase et agoniste des récepteurs 5‐HT4, aujourd’hui candidat préclinique d’intérêt potentiel dans le traitement de la maladie d’Alzheimer. De nombreux autres aspects de la polypharmacologie sont aujourd’hui à l’étude dans l’unité.
Lesinteractionsprotéine‐protéineLes programmes dévolus à la recherche de perturbateurs des interactions protéine‐protéine (IPPs) ont permis de mettre en évidence la pertinence de notre approche intégrant les activités de chimie médicinale et de chémoinformatique au service de la conception d’oligomères (hétéro)aromatiques comme mimes d’hélice alpha. La découverte du Pyridoclax, comme inhibiteur de la protéine anti‐apoptotique Mcl‐1, motive l’unité à élargir les objectifs scientifiques concernant la mise en place d’un environnement permettant d’optimiser la définition de nouveaux châssis moléculaires ciblant la perturbation des IPPs (plateforme de Drug Design orientée IPPs). L’intérêt de la construction de tels objets réside dans l’éventualité de reproduire des fonctions chimiques de protéines (mimes d’hélices alpha, mimes de feuillets bêta) impliquées dans des processus physiologiques ou/et pathologiques. Le savoir‐faire acquis dans l’unité est aujourd’hui exploité dans plusieurs programmes innovants et notamment en cancérologie.

Missions : Développer des outils de diagnostic pour l’imagerie TEP
Les activités de recherche combinent à la fois une recherche fondamentale de développement de nouveaux outils en chimie/radiochimie et d’investigation de nouveaux biomarqueurs moléculaireset une recherche finalisée avec des objectifs très précis en termes d’agents d’imagerie (TEP et multimodaux) pour l’oncologie
Compétences
Les travaux se situent à l’interface de la chimie et la biologie, et se caractérisent par un aspect très fortement multidisciplinaire impliquant toute une chaîne de compétences : 1) chimie (synthèse de ligands cible et de précurseur de marquage), 2) radiochimie du carbone-11, fluor-18 ou gallium 68 (étude de radiomarquage et mise au point de radiosynthèse), 3) automatisation (production sur module de synthèse des radiotraceurs destinés à être évalués in vivo), 4) évaluation in vitro, ex vivo et in vivo chez l’animal (autoradiographie, imagerie microTEP, microdissection), 5) études de métabolisation et radiométabolisation et, 6) si approprié, élaboration des dossiers DME (Dossier du Médicament Expérimental) pour les exploitations TEP cliniques.

In2010,theCOBRAlaboratorychosetoputasynthesiscenterinplace(C2iOrga)whichoffersawidevarietyofexpertiseintheareasofheterocyclesynthesis,fluorophores,bioconjugates,andfluorinated biomolecules. This center allows for internal response to different problems ofsynthesis linked to the chemistry‐biology research programs, while assuring several generalmissionsofthegroup(contributingtotheNationalChemicalLibrary,promotingandsupportingthe fundamental results of the group, conducting the exemplification of patents). An externalactivityaimsatansweringtothedifferentdemandsofbiologistsandpharmaceuticalpartners,andhelpsindevelopingindustrialpartnerships.

However, theselectivedestructionofcancercellswithoutaffectinghealthytissuesemergedasoneofthemajorissuesincancerchemotherapy.Improvingthedrugmonitoringanddeliveryisthereforehighlydesirablebutitremains a significant challenge despite recent improvements. Consequently, future collaborationsbetween our teams will aim at developing an effective SN38‐based prodrug bearing the urotensin 2subunitastargetingligand,andaself‐immolativelinkertoreleasetheantitumoragentselectivelyinsidetumor cells. A first strategywill rely on the use of a linker based on disulfide bonds that permits thereleaseofthetherapeuticagentSN38inthepresenceofintracellularbiothiolsthatarepresentincancercells. In an alternative strategy, the drug release could be triggered only in the acidic environment ofcancer cells. To this end, different well‐established pH‐dependence self‐immolable linkers could beenvisioned such as imines, enol ethers acetals, or an aromatization process of the oxabicycle into thecorresponding azaphthalimide, a reaction recently developed in the Bio‐organic team in collaborationwiththegroupofDr.H.Castel(Jouannoetal.JOrgChem2014,1303–1319).

TotalsynthesisofazaphilonesChaetomugilins are natural products isolated from a strain of Chaetomiumglobosum originally isolatedfromthemarinefishMugilcephalus,belongingtotheazaphilonefamily.Theseazaphilonesaresecondarymetabolites thathave theability to trapproteins irreversiblyvia their reactionwithprimaryaminesofaminoacids (mainly lysines); thisability is certainlyat theoriginof theirbiologicalactivity. Indeed, inadditiontotheiroriginalstructure,thesenaturalproductshavesignificantcytotoxicityagainstapanelof39 human cancer cell lines with log GI50 values between ‐5 and ‐6 (J. Antibiot. 2008, 61, 615‐622;Tetrahedron2009,65,7580‐7586).Thesynthesisofchaetomugilinsandrelatedchaetoviridinsisinprogress in the laboratory. Structure‐activity relationship and protein target fishing will also beundertaken.

Présentation de l’unité de recherche
L'activité de recherche du LG2A se situe essentiellement dans la glycochimie : le développement de voies d’accès à des molécules d’intérêt issues de mono‐ et oligosaccharides par synthèse chimique ou par dépolymérisation de polysaccharides suivie de fonctionnalisation. Nous sommes structurés en 2 axes :
Le premier se situe dans le domaine de la Chimie pour le vivant, avec les composantes principales suivantes : les Antimicrobiens (antiviraux, antibactériens, antiparasitaires) et Anticancéreux, et la Reconnaissance Moléculaire et Vectorisation (interactions sucres‐lectines, multivalence, capteurs, oligosaccharides). Nous avons des compétences largement reconnues aux niveaux nationaux et internationaux.
Le deuxième axe est celui de la Chimie pour le Développement Durable, comprenant l’Utilisation et Développement de Méthodologies en chimie durable appliquée aux substrats glucidiques et la Valorisation de la Biomasse, et notamment des agro‐ressources. Nous développons des méthodologies de synthèse innovantes à partir des agro‐ressources.
Le Laboratoire est placé nœud d'un réseau dense de relations avec des organismes de recherche, des industries, et des réseaux (IEED Pivert, RS2E, EPNOE).
