INTRODUCTION ET OBJET DU TEST La gélose Granada™ est un milieu sélectif utilisé pour la détection et l’identification directe de Streptococcus agalactiae chez les femmes enceintes et chez les nouveaux nés à partir de prélèvements d’origine clinique (1, 2, 3 et 4). Le milieu a été décrit par le Dr De La Rosa et al .Il a été développé selon les premiers travaux de A.K.M.S. Islam (8) et du Dr de la Rosa (9 , 10).
Les S. agalactiae sont responsables d’infections graves chez le nouveau né (méningite). Leur détection est particulièrement importante pour la prévention, le traitement et le suivi des infections.
PRINCIPELe milieu Granada est basé sur la formule originale décrite par le Dr De La Rosa :- La gélose Granada est constituée d’une base nutritive
associant différentes peptones, du pyruvate et du glucose.
- la présence de méthotrexate, de sérum de cheval et d’amidon permet la production d’un pigment orange à rouge spécifique des colonies de streptocoques du groupe B hémolytiques (5).
- Le mélange d'antibiotiques inhibe la plupart des bactéries gram négatif et des levures .

CONDITIONS DE STOCKAGE • Les boîtes se conservent entre 2°C et 8°C dans leur
coffret jusqu’à la date de péremption.• La durée de conservation des boîtes hors du coffret,
en sachet cellophane, est de 2 semaines à 2-8°C.
ECHANTILLONS Les échantillons sont constitués de prélèvements d’origine ano- vaginale et urinaire chez la femme enceinte ou liquides gastriques ingérés du nouveau né. Ils sont directement ensemencés sur la gélose ou après enrichissement en bouillon Todd Hewitt + antibiotiques. Il convient de respecter les bonnes pratiques de prélèvement et de transport, adaptées à chaque type de prélèvement.

MODE OPERATOIRE La gélose Granada™ peut être utilisée selon deux protocoles : - avec enrichissement du prélèvement en bouillon sélectif :
TODD HEWITT + antibiotiques (1, 7). Et / ou- ensemencement direct du prélèvement (7).
1. Laisser les boîtes revenir à température ambiante.2. Ensemencer le prélèvement. 3. L’incubation est réalisée en anaérobiose à
37°C pendant 18 à 24 heures. Après la première lecture, l'incubation doit être prolongée de 24 heures supplémentaires en cas d'absence de colonies caractéristiques.
Remarque:L’enrichissement augmente significativement la sensibilité du test (7) .
Les performances ont été obtenues en suivant la méthodologie et une température d'incubation de 37°C Tout autre choix de température d'incubation est de la responsabilité de l'utilisateur pour vérifier que les performances sont maintenues.
LECTURE ET INTERPRETATION • Après incubation, observer la croissance bactérienne
et l’aspect des colonies : les colonies caractéristiques de Streptococcus agalactiae sont oranges à rouges.
• Les colonies de micro-organismes appartenant à d’autres espèces sont soit inhibées, soit se développent en donnant une autre couleur.
CONTROLE DE QUALITE
Protocole : La fertilité et la sélectivité du milieu peut être testée vis-à-vis des souches suivantes:
• Streptococcus agalactiae ATCC® 12386. • Enterococcus faecalis ATCC® 29212• Escherichia coli ATCC 25922.

Remarque : Il est de la responsabilité de l'utilisateur de prendre en compte la nature de l'application et la législation locale en vigueur pour la mise en oeuvre du contrôle de qualité (fréquence, nombre de souches, température d'incubation).
LIMITES DU TEST• Le développement est fonction des exigences propres à
chaque micro-organisme. Il est donc possible que certaines souches de S. agalactiae ayant des exigences spécifiques (substrat, température, atmosphère d’incubation….) ne se développent pas.
• En fonction des prélèvements analysés il est possible d’associer une gélose au sang non sélective.
• Certaines souches de S.agalactiae ne donnent pas de colonies caractéristiques en particulier les souches non hémolytiques (2,3 %).(6)
• Certaines souches résistantes aux antibiotiques présents dans le milieu peuvent se développer sans donner des colonies caractéristiques.
• La coloration orange à rouge présente différentes intensités selon la souche.
• En cas de réalisation d'un test complémentaire d'identification à partir de colonies isolées sur la gélose Granada™, les réactifs VITEK® GPI (Réf. V 1305) et VITEK® 2 GP (Réf. 21 342) ne sont pas recommandés.
PERFORMANCES Les performances ont été évaluées à 37°C sur 55 souches : 26 souches hémolytiques de S.agalactiae, 5 souches non hémolytiques de S.agalactiae,6 Streptococcus / Enterococcus et 18 souches de différentes espèces. Les souches ont été ensemencées: - directement sur la gélose Granada - et après enrichissement pendant une nuit en bouillon TODD
HEWITT + antibiotiques . Après 18 à 42 heures d'incubation à 37°C, en atmosphère anaérobie, les colonies caractéristiques ont été observées.

ELIMINATION DES DECHETS

SUMMARY AND EXPLANATION Granada™ Agar is a selective medium for the screening and direct identification of Streptococcus agalactiaecarriage in pregnant women and newborns using clinical specimens (1, 2 , 3 , 4).The medium was first described by Dr De La Rosa and al.It was developed from the previous work of A.K.M.S Islam (8) and Dr De La Rosa (9, 10). S. agalactiae are responsible for serious infections in newborns (meningitis). Their detection is particularly important for the prevention, treatment, and monitoring of infections .
PRINCIPLEGranada Agar is based on the original formula described by Dr De La Rosa: - Granada Agar consists of a nutritive base combining
different peptones, pyruvate and glucose. - The methotrexate, horse serum and starch present in
the medium enable the production of an orange to red pigment which is characteristic of group B streptococcus colonies.(5)
- The antibiotic mixture inhibits most Gram-negative bacteria and yeasts.

WARNINGS AND PRECAUTIONS • For in vitro diagnostic use only. • For professional use only. • This kit contains products of animal origin. Certified
knowledge of the origin and/or sanitary state of the animals does not totally guarantee the absence of transmissible pathogenic agents. It is therefore recommended that these products be treated as potentially infectious, and handled observing the usual safety precautions (do not ingest or inhale).
• All specimens, microbial cultures and inoculated products should be considered infectious and handled appropriately. Aseptic technique and usual precautions for handling the bacterial group studied should be observed throughout this procedure. Refer to "CLSI®
M29-A, Protection of Laboratory Workers from Occupationally Acquired Infections; Approved Guideline – Current Revision". For additional information on handling precautions, refer to "Biosafety in Microbiological and Biomedical Laboratories – CDC/NIH – Latest edition", or the current regulations in the country of use.
• Do not use reagents after the expiry date. • Do not use reagents if the packaging is damaged. • Do not use contaminated plates or plates that exude
moisture.• Interpretation of the test results should be made taking
into consideration the patient's history, the source of the specimen, colonial and microscopic morphology and, if necessary, the results of any other tests performed.
• Use of the medium may be difficult for people who have problems recognizing colors.
• Performance data presented were obtained using the procedure and the incubation temperature indicated in this package insert. Any change or modification in the procedure may affect the results.
STORAGE CONDITIONS • Store the plates in their box at 2-8°C until the expiry
date.• If not in the box, plates can be stored in the cellophane
sachet for 2 weeks at 2-8°C.
SPECIMENSSpecimens consist of anovaginal and urine samples from pregnant women, or ingested gastric fluid from newborns. The samples are inoculated directly on the agar or following enrichment in Todd Hewitt Broth + Antibiotics. Good laboratory practices for collection and transport should be respected and adapted to the type of specimen.

INSTRUCTIONS FOR USE Granada™ Agar can be used according to two protocols: - enrichment of the specimen in selective broth: Todd
Hewitt + Antibiotics (1, 7). And / or - direct inoculation of the specimen (7).
1. Allow the plates to come to room temperature. 2. Inoculate the specimen. 3. Incubate at 37°C for 18-24 hours in anaerobic
conditions.After the first reading, incubation can be prolonged for an additional 24 hours if no typical colonies are observed.
Note: Enrichment significantly increases the sensitivity of the test (7).Performance data presented were obtained using the procedure and the 37°C incubation temperature. The user is responsible for any other choice of incubation temperature, to ensure that good performance is maintained.
READING AND INTERPRETATION • After incubation, observe the bacterial growth and the
appearance of the colonies: typical Streptococcus agalactiae colonies are orange to red.
• The growth of micro-organisms belonging to other species is either inhibited or the colonies produced are a different color.

Note:It is the responsibility of the user to perform Quality Control taking into consideration the intended use of the medium, and in accordance with any local applicable regulations (frequency, number of strains, incubation temperature etc.).
LIMITATIONS OF THE METHOD • Growth depends on the requirements of each individual
micro-organism. It is therefore possible that certain strains of S. agalactiae which have specific requirements (substrate, temperature, incubation conditions, etc.) may not develop.
• According to the specimens analyzed, a non-selective blood agar may be used in conjunction.
• Some strains of S.agalactiae may not produce typical colonies, particularly non-hemolytic strains (2.3 %) (6).
• Some strains which are resistant to the antibiotics in the medium may develop without producing typical colonies.
• The intensity of the orange to red color may vary depending on the strain.
• If a complementary identification test is to be performed using colonies isolated on Granada Agar, it is not recommended to use the VITEK® GPI (Ref. V 1305) and VITEK® 2 GP (Ref. 21 342) reagents.
PERFORMANCE Performance was evaluated at 37°C using 55 strains: 26 hemolytic S.agalactiae strains, 5 non-hemolytic S.agalactiae strains, 6 Streptococcus / Enterococcus and 18 strains from different species. The strains were inoculated: - directly on Granada agar - and following overnight enrichment in TODD HEWITT
+ antibiotics broth. After 18-42 hours of incubation at 37°C in anaerobic conditions, typical colonies were observed.

EINFÜHRUNG UND PRODUKTERKLÄRUNG Granada™ Agar ist ein Selektivmedium zur Anzucht und direkten Identifizierung von Streptococcus agalactiae bei schwangeren Frauen und Neugeborenen aus klinischen Proben (1, 2, 3, 4). Das Medium wurde erstmals von Dr. De La Rosa et al. beschrieben.Die Entwicklung des Mediums basiert auf den Arbeiten des A.K.M.S. Islam (8) und Dr. De La Rosa (9, 10).
S. agalactiae ist für schwere Infektionen bei Neugeborenen verantwortlich (Meningitis). Ihr Nachweis ist für die Prävention, Behandlung und Verlaufskontrolle der Infektionen besonders wichtig.
PRINZIPDas Granada Medium basiert auf der von Dr. De La Rosa beschriebenen Zusammensetzung: - Der Granada Agar besteht aus einer Nährstoffbasis aus
verschiedenen Peptonen, Pyruvat und Glukose. - Die Anwesenheit von Methotrexat, Pferdeserum und
Stärke ermöglicht die Bildung eines orange- bis rot-farbenen Pigmentes, das für Kolonien von Streptokokken der Gruppe B charakteristisch ist (5).

VORSICHTSMASSNAHMEN Nur für die in vitro Diagnostik. Nur für die Verwendung durch Fachkundige bestimmt. Dieser Kit enthält Bestandteile tierischen Ursprungs. Da durch die Kontrolle der Herkunft und/oder des Gesundheitszustandes der Tiere nicht völlig gewähr-leistet werden kann, dass diese Produkte keine übertragbaren pathogenen Agenzien enthalten, ist es empfehlenswert, diese als potenziell infektiös zu betrachten und unter Beachtung entsprechender Vorsichtsmaßnahmen zu behandeln (nicht einnehmen, nicht einatmen). Die Proben, Mikroorganismen und beimpften Produkte müssen als potenziell infektiös betrachtet und unter Beachtung geeigneter Vorsichtsmaßnahmen sach-gemäß behandelt werden. Während der gesamten Testdurchführung müssen aseptische Arbeitsbedingun-gen und entsprechende Vorsichtsmaßnahmen für die zu untersuchende Keimgruppe eingehalten werden, siehe „CLSI® M29-A, Protection of Laboratory Workers from Occupationally Acquired Infections, Approved Guideline –aktuelle Revision“. Die Platten nach Ablauf des Verfallsdatums nicht mehr verwenden. Platten mit beschädigter Verpackung nicht verwenden. Kontaminierte oder eingetrocknete Platten nicht verwenden. Bei der Interpretation der Ergebnisse müssen der klinische Kontext, die Herkunft der Probe, die Kolonie- und mikroskopische Morphologie und gegebenenfalls die Ergebnisse anderer Tests berücksichtigt werden. Der Gebrauch des Mediums ist für Personen mit Problemen bei der Farbwahrnehmung möglicherweise schwierig. Die angegebenen Leistungsdaten wurden gemäß der vorliegenden Gebrauchsanweisung und der in dieser Packungsbeilage angegebenen Inkubationstemperatur ermittelt. Jede Abweichung von diesem Verfahren kann die Ergebnisse beeinflussen.
LAGERUNGSBEDINGUNGEN Die Platten sind in ihrem Originalkarton bei 2-8°C bis zum Verfallsdatum haltbar. Außerhalb des Originalkartons beträgt die Haltbarkeit der Platten im Zellophanbeutel 2 Wochen bei 2-8°C.
PROBENEs können anovaginale Abstriche sowie Urinproben von schwangeren Frauen oder aspirierter Magensaft von Neugeborenen untersucht werden. Die Proben werden direkt oder nach Anreicherung in Todd Hewitt Bouillon mit Antibiotika auf den Agar überimpft. Bei der Gewinnung und dem Transport der Proben sollten die GLP-Richtlinien beachtet und auf das jeweilige Untersuchungsmaterial abgestimmt werden.

GEBRAUCHSANWEISUNG Granada™ Agar kann gemäß 2 Verfahren verwendet werden: - nach Anreicherung der Probe in einer Selektivbouillon:
Todd Hewitt mit Antibiotika (1, 7) und / oder
- direkte Beimpfung mit der Probe (7).
1. Bringen Sie die Platten vor Gebrauch auf Raumtemperatur.
2. Überimpfen Sie die Probe. 3. Inkubieren Sie die Platte unter anaeroben Bedingungen
bei 37°C für 18 bis 24 h. Wenn nach der ersten Ablesung keine charakteristischen Kolonien sichtbar sind, verlängern Sie die Inkubation um weitere 24 h. Anmerkung: Durch vorherige Anreicherung wird die Sensitivität des Tests signifikant erhöht (7).
Die Leistungsdaten wurden gemäß der vorliegenden Gebrauchsanweisung und einer Inkubations-temperatur von 37°C ermittelt. Die Wahl einer anderen Inkubationstemperatur liegt in der Verantwortung des Anwenders, der sich vergewissern muss, dass die Leistungsdaten dadurch nicht beeinträchtigt werden.
ABLESUNG UND INTERPRETATION Nach der Inkubation das Keimwachstum und das Aussehen der Kolonien beurteilen: charakteristische Streptococcus agalactiae Kolonien sind orangefarben bis rot. Andere Spezies werden entweder gehemmt oder bilden Kolonien mit anderer Farbe.

Hinweis:
Es liegt in der Verantwortung des Anwenders, die Qualitätskontrolle unter Berücksichtigung des Verwendungszweckes und in Übereinstimmung mit den regional gültigen Vorschriften (Frequenz, Anzahl der Stämme, Inkubationstemperatur...) durchzuführen.
LIMITIERUNGENDas Wachstum hängt von den Wachstumsansprüchen des jeweiligen Keimes ab. Es ist deshalb möglich, dass einige S.agalactiae Stämme mit besonderen Wachstumsansprüchen (Substrat, Temperatur, Inkubationsatmosphäre...) nicht wachsen. Je nach Untersuchungsmaterial kann der Agar zusammen mit einem nicht selektiven Blutagar verwendet werden. Einige S.agalactiae, insbesondere nicht hämolysierende Stämme, bilden keine charakteristischen Kolonien (2,3%) (6). Einige Stämme, die gegen die im Agar enthaltenen Antibiotika resistent sind, können wachsen, ohne charakteristische Kolonien zu bilden. Je nach Stamm ist die orange-farbene bis rote Färbung von unterschiedlicher Intensität. Wenn ein weiterer Identifizierungstest mit Kolonien durchgeführt wird, die auf Granada™ Agar isoliert wurden, ist es nicht empfehlenswert, die Reagenzien VITEK® GPI (Best.Nr. V1305) und VITEK® 2 GP (Best.Nr. 21 342) zu verwenden.
PERFORMANCE Die Leistungsdaten wurden bei 37°C mit 55 Stämmen evaluiert:26 hämolysierende S.agalactiae Stämme, 5 nicht hämolysierendeS.agalactiae Stämme, 6 Streptococcus / Enterococcus und 18 Stämme von anderen Spezies. Die Stämme wurden überimpft: - direkt auf den Granada Agar - und nach Anreicherung für eine Nacht in TODD HEWITT
Bouillon mit Antibiotika. Nach 18 bis 42 h Inkubation bei 37°C in anaerober Atmosphäre wurden folgende charakteristischen Kolonien beobachtet:

INTRODUCCIÓN Y OBJETO DEL ANALISIS Agar Medio Granada™ es un medio selectivo para la selección e identificación directa de Streptococcus agalactiae portado por las mujeres embarazadas y los recién nacidos a partir de muestras clínicas (1, 2, 3, 4). El Dr. de la Rosa et al fueron los primeros en descubrir el medio. Se desarrolló según los trabajos previos de A.K.M.S Islam (8) y del Dr. de la Rosa (9, 10). S. agalactiae es responsable de infecciones graves en los recién nacidos (meningitis). Su detección es particularmente importante para la prevención, tratamiento, y seguimiento de las infecciones.
PRINCIPIOAgar Medio Granada se basa en la fórmula original descrita por el Dr. de la Rosa: - Agar Medio Granada consiste en una base nutritiva
que combina diferentes peptonas, piruvato y glucosa. - El metotrexato, suero de caballo y almidón presente
en el medio permiten la producción de un pigmento de color entre naranja y rojo que es característico de grupos de colonias de estreptococos B. (5)
- La mezcla antibiótica inhibe la mayoría de las bacterias gram negativas y las levaduras.

PRECAUCIONES DE UTILIZACIÓN Para diagnóstico in vitro exclusivamente. Exclusivamente para uso profesional. Este envase contiene compuestos de origen animal. La falta de control sobre el origen y/o el estado sanitario de los animales, no nos permite garantizar de forma absoluta que estos productos no contengan algún agente patógeno transmisible, por lo que se recomienda manipularlos mediante las precauciones de utilización relativas a los productos potencialmente infecciosos: (no ingerir, ni inhalar). Todas las muestras, cultivos microbianos y productos inoculados deben considerarse como potencialmente infecciosos y se manipularán de un modo apropiado. Durante la manipulación, se respetarán las técnicas asépticas y las precauciones usuales de manipulación para el grupo bacteriano estudiado; consultar: "CLSI® M29-A, Protection of Laboratory Workers from Occupationally Acquired Infections; Approved Guideline – Revisión en vigor". Para obtener información complementaria sobre las precauciones de manipulación, consultar "Biosafety in Microbiological and Biomedical Laboratories – CDC/NIH – Última edición", o la reglamentación en vigor en el país de utilización. No usar los reactivos pasada su fecha de caducidad. No usar los reactivos si el envase está deteriorado. No utilizar placas contaminadas o las que exudan humedad. Se deben interpretar los resultados de las pruebas teniendo en cuenta el historial clínico del paciente, el origen de la muestra, la morfología colonial y microscópica y, en su caso, los resultados de cualquier otra prueba que se haya realizado. El uso del medio podría resultar difícil para alguna persona que tenga problemas en reconocer los colores. Los resultados presentados se obtuvieron mediante el procedimiento y a la temperatura de incubación indicada en esta ficha técnica. Cualquier cambio o modificación en el procedimiento podría alterar los resultados.
CONDICIONES DE CONSERVACIÓN Almacenar las placas en su envase a 2-8°C hasta su fecha de caducidad. Al no estar en su envase, se pueden conservar las placas en su bolsita de celofán durante 2 semanas a 2-8°C.
MUESTRAS Las muestras consisten en muestras anovaginales y de orina de mujeres embarazadas, o el fluido gástrico ingerido de los recién nacidos.Se siembran directamente sobre el agar o tras el enriquecimiento en caldo Todd Hewitt + Antibióticos.Se deben respetar las buenas prácticas de laboratorio para la recogida y transporte, adaptándolas al tipo de muestra.

TÉCNICA Se puede usar Agar Medio Granada™ según dos protocolos: - enriquecimiento de la muestra en caldo selectivo:
Todd Hewitt + Antibióticos (1, 7). Y / o - inoculación directa de la muestra (7).
1. Permitir que las placas alcancen la temperatura ambiente.
2. Sembrar la muestra. 3. Incubar a 37°C durante 18-24 horas en condiciones
anaeróbicas.Tras la primera lectura, la incubación se puede prolongar otras 24 horas si no se observan colonias típicas .
Nota: El enriquecimiento aumenta considerablemente la sensibilidad de la prueba (7).
Los resultados presentados se obtuvieron mediante el procedimiento indicado y con una temperatura de incubación de 37°C. El usuario es responsable de cualquier otra elección de la temperatura de incubación, para garantizar un buen funcionamiento del método.
LECTURA E INTERPRETACIÓN Después de la incubación, observar el crecimiento bacteriano y el aspecto de las colonias: las colonias de Streptococcus agalactiae son típicamente de color naranja a rojo. El crecimiento de los microorganismos pertenecientes a otras especies se inhibe o las colonias producidas son de un color diferente.

Nota:Es responsabilidad del usuario llevar a cabo el Control de Calidad teniendo en cuenta el uso adecuado del medio, y conforme a cualquier regulación local aplicable (frecuencia, número de cepas, temperatura de incubación, etc.).
LÍMITES DE LA PRUEBA El crecimiento depende de los requisitos de cada microorganismo individual. Es por tanto posible que ciertas cepas de S. agalactiae que tengan requisitos específicos (sustrato, temperatura, condiciones de incubación, etc.) puedan no desarrollarse. Según las muestras analizadas, se puede utilizar un agar sangre no selectivo al mismo tiempo. Algunas cepas de S.agalactiae pueden no producir colonias típicas, en particular cepas no hemolíticas ( 2,3%) (6). Algunas cepas resistentes a los antibióticos del medio pueden desarrollarse sin producir colonias típicas. El color naranja a rojo puede presentar una intensidad diferente dependiendo de la cepa. Si se hace una identificación complementaria usando colonias aisladas en medio Granada, no se recomienda usar VITEK® GPI ( Ref. V 1305) y VITEK®
2 GP ( Ref . 21342).
PRESTACIONES Las prestaciones se evaluaron a 37°C usando 55 colonias:26 de S.agalactiae hemolítico, 5 de S.agalactiae no-hemolítico, 6 Streptococcus / Enterococcus y 18 cadenas de diferentes especies Se inocularon las colonias: - Directamente de medio Granada - Y tras un enriquecimiento nocturno en caldo TODD
HEWITT + antibióticos. Tras 18-42 hours de incubación a 37°C en condiciones anaeróbicas, se observaron colonias típicas.

INTRODUZIONE E OBIETTIVO DEL TEST L’agar Granada™ è un terreno selettivo utilizzato per la ricerca e l’identificazione diretta di Streptococcus agalactiae nelle donne in gravidanza e nei neonati partendo da prelievi di origine clinica (1, 2, 3 et 4). Il terreno è stato descritto dal Dr. De La Rosa e al.E’ stato sviluppato secondo i primi lavori di A.K.M.S. Islam (8) e del Dr. De la Rosa (9 , 10).
Gli S. agalactiae sono responsabili di gravi infezioni nel neonato (meningiti). La loro ricerca è particolarmente importante per la prevenzione, la terapia ed il monitoraggio delle infezioni.
PRINCIPIOIl terreno Granada™si basa sulla formula originale descritta dal Dr. De La Rosa :- L’agar Granada™è costituito da una base nutritiva che
associa diversi peptoni, piruvato e glucosio. - La presenza di metotrexato, di siero di cavallo e di
amido, permette la produzione di un pigmento dall’arancione al rosso specifico delle colonie degli streptococchi emolitici di gruppo B (5).
- La miscela di antibiotici inibisce la maggior parte dei batteri gram negativi e dei lieviti.

tenendo conto del contesto clinico, dell’origine del prelievo, degli aspetti macro e microscopici e, eventualmente, dei risultati di altri test.
• L’utilizzazione del terreno può presentare dei problemi per le persone che hanno difficoltà nel riconoscere i colori.
• Le performance riportate sono state ottenute con la metodologia e la temperatura di incubazione indicate in questa scheda tecnica. Qualsiasi deviazione dal procedimento indicato può alterare i risultati.
CONDIZIONI DI CONSERVAZIONE • Le piastre vanno conservate tra 2°C e 8°C nella
confezione fino alla data di scadenza.• Fuori della confezione, le piastre si conservano per
2 settimane a 2-8°C nel loro sacchetto di cellophane.
CAMPIONI I campioni sono costituiti da prelievi d’origine ano-vaginale ed urinaria nelle donne in gravidanza o da liquidi gastrici ingeriti dal neonato. Vanno seminati sull’agar direttamente o dopo arricchimento in brodo Todd Hewitt + antibiotici. Per il prelievo ed il trasporto, si devono rispettare le buone pratiche di laboratorio adattate ad ogni tipo di prelievo.

PROCEDIMENTOL’agar Granada™ può essere utilizzato con 2 diversi protocolli : - Con arricchimento del prelievo in brodo selettivo :
TODD HEWITT + antibiotici (1, 7). E / o- semina diretta del prelievo (7).
1. Portare le piastre a temperatura ambiente.2. Seminare il prelievo.
3. L’incubazione va eseguita in anaerobiosi a 37°C per 18-24 ore. Dopo la prima lettura, in caso di assenza di colonie caratteristiche, l’incubazione deve essere prolungata di 24 ore.
Nota: L’arricchimento aumenta significativamente la sensibilità del test (7).
Le performance sono state ottenute seguendo la metodologia indicata e con una temperatura di incubazione di 37°C. Ogni altra scelta di temperatura di incubazione è sotto la responsabilità dell’utilizzatore che deve verificare che le performance siano mantenute.
LETTURA E INTERPRETAZIONE • Dopo l’incubazione, osservare la crescita batterica e
l’aspetto delle colonie : le colonie caratteristiche di Streptococcus agalactiae sono di colore da arancione a rosso.
• Le colonie di microrganismi appartenenti ad altre specie, o vengono inibite, o si sviluppano dando un colore diverso.

Protocollo : La fertilità e la selettività del terreno possono essere testate nei confronti dei seguenti ceppi: • Streptococcus agalactiae ATCC® 12386. • Enterococcus faecalis ATCC® 29212• Escherichia coli ATCC 25922.

E’ responsabilità dell’utilizzatore assicurarsi che il controllo di qualità corrisponda a quanto previsto dalla legislazione vigente tenendo in considerazione la natura dell’applicazione (frequenza, numero di ceppi, temperatura di incubazione). .
LIMITI DEL METODO • La crescita dipende dalle particolari esigenze di
ciascun microrganismo. E’ quindi possibile che alcuni ceppi di S. agalactiae con esigenze specifiche (substrato, temperatura, atmosfera d’incubazione...)non siano in grado di svilupparsi.
• In funzione dei prelievi analizzati è possibile associare un agar al sangue non selettivo.
• Alcuni ceppi di S. agalactiae non danno colonie caratteristiche, in particolare i ceppi non emolitici (2,3 %) (6).
• Alcuni ceppi resistenti agli antibiotici presenti nel terreno possono svilupparsi senza dare colonie caratteristiche.
• La colorazione arancione può presentare intensità differenti a seconda del ceppo.
• In caso di esecuzione di un test di identificazione complementare partendo da colonie isolate sull’agar Granada™, non sono raccomandati i reattivi VITEK®
GPI (Cod. V 1305) e VITEK® 2 GP (Cod. 21 342)
PERFORMANCE Le performance sono state valutate a 37°C su 55 ceppi: 26 ceppi emolitici di S. agalactiae, 5 ceppi non emolitici diS. agalactiae, 6 Streptococchi / Enterococchi e 18 ceppi de differenti specie. I ceppi sono stati seminati: - Direttamente sull’agar Granada™ - E dopo arricchimento per una notte in brodo TODD
HEWITT + antibiotici. Dopo 18 - 42 ore di incubazione a 37°C, in anaerobiosi, sono state osservate le colonie caratteristiche.

INTRODUÇÃO E OBJECTIVO DO TESTE A gelose Granada é um meio selectivo utilizado para a detecção e a identificação directa de Streptococcus agalactiae nas mulheres grávidas e nos recém-nascidos a partir de colheitas/coletas de origem clínica (1, 2, 3 e 4). O meio foi descrito por Dr De La Rosa et al .Foi descrito segundo os primeiros trabalhos de A.K.M.S. Islam.(8) e do Dr de la Rosa (9 , 10).
Os S. agalactiae são responsáveis por infecções graves no recém-nascido (meningite). A sua detecção é particularmente importante para a prevenção, tratamento e seguimento das infecções.
PRINCÍPIOO meio Granada baseia-se na fórmula original descrita pelo Dr De La Rosa : - A gelose Granada é constituída por uma base nutritiva
que associa diferentes peptonas, piruvato e glucose. - A presença de metotrexato, soro de cavalo e amIdo
permite a produção de um pigmento cor de laranja a vermelho específico das colónias de estreptotocos do grupo B hemolíticos (5).
- A mistura de antibióticos inibe a maioria das bactérias gram negativas e leveduras.

efectuada tendo em conta o contexto clínico, a origem da amostra, os aspectos macro e microscópicos e, eventualmente, os resultados de outros testes.
• A utilização do meio pode ser delicada para pessoas que tenham dificuldades na apreciação de cores.
• O comportamento funcional apresentado foi obtido com o procedimento e a temperatura de incubação indicados no folheto informativo. Qualquer desvio de procedimento pode alterar os resultados.
CONDIÇÕES DE ARMAZENAMENTO • As placas conservam-se entre 2°C e 8°C dentro da
embalagem até à data de validade.• O tempo de conservação das placas fora da
embalagem, em saqueta/sachet de celofane, é de 2 semanas a 2° - 8° C.
AMOSTRAS As amostras são constituídas de colheitas/coletas ano-vaginais e urinárias da mulher grávida ou líquidos gástricos ingeridos do recém-nascido. São directamente semeadas na gelose ou após o enriquecimento em caldo Todd Hewitt + antibióticos. É conveniente respeitar as boas práticas de colheita/coleta e de transporte, adaptadas a cada tipo de colheita/coleta.

PROCEDIMENTOA gelose Granada pode ser utilizada em conformidade com dois protocolos: - Com enriquecimento da colheita/coleta em caldo
selectivo: TODD HEWITT + antibióticos (1, 7). E / ou- Sementeira directa da amostra (7).
1. Deixar as placas atingir a temperatura ambiente.2. Semear a amostra.
3. A incubação é efectuada em anaerobiose a 37°C durante 18 a 24 horas. Após a primeira leitura, a incubação deve ser prolongada 24 horas suplementares em caso de ausência de colónias características. Nota: O enriquecimento aumenta significativamente a sensibilidade do teste (7).
O comportamento funcional foi obtido seguindo o procedimento e uma temperatura de incubação de 37°C. Qualquer outra escolha de temperatura é da responsabilidade do utilizador para verificar que o comportamento funcional é mantido.
LEITURA E INTERPRETAÇÃO • Após incubação, observar o crescimento bacteriano e o
aspecto das colónias: as colónias características de Streptococcus agalactiae são laranja a vermelhas.
• As colónias de microrganismos pertencentes a outras espécies são inibidas ou desenvolvem-se dando outra cor.
CONTROLO DE QUALIDADE
Protocolo: A fertilidade e a selectividade do meio pode ser testada em relação às estirpes/cepas seguintes:
• Streptococcus agalactiae ATCC® 12386. • Enterococcus faecalis ATCC® 29212 • Escherichia coli ATCC 25922.

Nota:É da responsabilidade do utilizador ter em conta a natureza da aplicação e a legislação local em vigor para a execução do controlo de qualidade (frequência, número de estirpes/cepas, temperatura de incubação... ).
LIMITES DO TESTE• O desenvolvimento depende das exigências específicas
de cada microrganismo. Portanto, é possível que algumas estirpes/cepas com exigências específicas (substrato, temperatura, atmosfera de incubação….) não se desenvolvam.
• Em função das amostras analisadas, é possível associar uma gelose de sangue não selectiva.
• Algumas estirpes/cepas de S.agalactiae não formam colónias características, em particular as estirpes/cepas não hemolíticas (2,3 %).(6)
• Algumas estirpes/cepas resistentes aos antibióticos presentes no meio podem desenvolver-se sem formar colónias características.
• A coloração laranja a vermelho apresenta diferentes intensidades consoante a estirpe/cepa.
• Em caso de realização de um teste complementar de identificação a partir de colónias isoladas em gelose Granada™, os reagentes VITEK® GPI (Ref. V 1305) e VITEK® 2 GP (Ref. 21 342) não são aconselháveis.
COMPORTAMENTO FUNCIONAL O comportamento funcional foi avaliado a 37°C com 55 estirpes/cepas: 26 estirpes/cepas hemolíticas de S.agalactiae, 5estirpes/cepas de S.agalactiae, 6 Streptococcus / Enterococcus e 18 estirpes/cepas de diferentes espécies. As estirpes/cepas foram semeadas: - Directamente em gelose Granada - e após enriquecimento durante uma noite em caldo
TODD HEWITT + antibióticos. Após 18 a 42 horas de incubação a 37°C, em atmosfera anaeróbia, foram observadas as colónias características.

SAMMANFATTNING OCH FÖRKLARING Granada™ Agar är ett selektivt medium för screening och direktidentifiering av Streptococcus agalactiae-förekomst i kliniska prov hos gravida kvinnor och nyfödda (1, 2 , 3 , 4). Mediet beskrevs ursprungligen av Dr De La Rosa et al.Det utvecklades med utgångspunkt i det tidigare arbetet av A.K.M.S Islam (8) och Dr De La Rosa (9, 10). S. agalactiae kan orsaka allvarliga infektioner hos nyfödda (meningit). Detektion av dessa är av stor betydelse för prevention, behandling samt övervakning av infektioner.
METODGranada Agar är baserad på den ursprungliga formeln beskriven av Dr De La Rosa : - Granada Agar består av en näringsbas som
kombinerar olika peptoner, pyruvat samt glukos - Metotrexat, hästserum och stärkelse som ingår i
mediet möjliggör produktion av ett orange-rött-pigment som är karakteristiskt för kolonier av grupp B streptokocker (5).
- Antibiotika-blandningen hämmar de flesta andra Gram-negativa bakterier och jäster.

avsöndrar fukt.• Tolkningen av testresultaten ska göras med hänsyn
till patientens anamnes, provkällan, kolonimorfologi och mikroskopisk morfologi och, om nödvändigt, resultat av andra utförda tester.
• Användning av mediet kan vara svår för personer med nedsatt färgseende.
• Data angående prestanda som presenteras har erhållits med hjälp av den metod samt inkuberingstemperatur som anges i denna bipacksedel. Varje ändring i utförandet kan påverka resultaten.
FÖRVARING • Förvara plattorna I sin låda vid 2-8°C fram till
utgångsdatum.• Utanför lådan kan plattorna förvaras i
cellofanpåsen i 2 veckor vid 2-8°C.
PROVERProvmaterial består av anovaginal samt urinprov från gravida kvinnor, eller intagen magsaft från nyfödda. Proven inokuleras direkt på agarplattan eller efter anrikning i Todd Hewitt-buljong + antibiotika. God laboratoriesed (GLP) ska respekteras vid insamling och transport och anpassas till varje provtyp.

BRUKSANVISNING Granada™ Agar kan användas enligt två protokoll: - Anrikning av proven i selektiv buljong: Todd Hewitt +
antibiotika (1, 7). Och/eller- Direktinokulering av proven (7).
1. Låt plattorna anta rumstemperatur.2. Inokulera proverna. 3. Inkubera vid 37°C i 18-24 timmar under anaeroba
förhållanden. Om inga typiska kolonier observeras efter första läsningen, förläng inkuberingen med ytterligare 24 timmar. Obs! Anrikning ökar testets känslighet avsevärt (7).
Data angående prestanda som presenteras har erhållits med hjälp av metoden samt inkuberingstempertaturen 37°C. Det är användarens ansvar att val av varje annan inkuberingstemperatur är lämplig med hänsyn till bibehållen god prestanda.
AVLÄSNING OCH TOLKNING • Studera bakterietillväxt och koloniernas utseende efter
inkubationen: typiska Streptococcus agalactiae-kolonier är orange-röda.
• Tillväxt av mikroorganismer tillhörande andra arter är antigen hämmad eller så har de producerade kolonierna en annan färg.

Obs:Det är användarens ansvar att utföra kvalitetskontroll med hänsyn till den planerade användningen av mediet och i enlighet med lokala tillämpliga förhållningsregler (frekvens, antal stammar, inkubationstemperatur etc.).
METODENS BEGRÄNSNINGAR • Tillväxt beror på behoven hos varje enskild
mikroorganism. Det är därför möjligt att vissa stammar av S. agalactiae som har specifika behov (substrat, temperatur, inkuberingsförhållden etc.) inte tillväxter.
• Beroende på de provtyper som analyseras kan även en icke-selektiv blodagar användas i förbindelse.
• Det är möjligt att vissa stammar av S.agalactiae inte producerar typiska kolonier, speciellt icke-hemolytiska stammar (2.3 %) (6).
• Vissa stammar som är resistenta mot antibiotikan i mediet kan komma att utvecklas utan att typiska kolonier produceras.
• Färgen orange kan presentera olika orange intensitet beroende på stammen.
• Om ett kompletterande identifieringstest skall utföras med hjälp av kolonier som isolerats på Granada Agar, är det inte att rekommendera att reagenserna VITEK® GPI (Art. nr. V 1305) och VITEK® 2 GP (Art.nr. 21 342) används.
PRESTANDA Prestanda utvärderades vid 37°C med hjälp av 55 stammar: 26 hemolytiska S.agalactiae-stammar, 5 icke-hemolytiska S.agalactiae-stammar, 6 Streptococcus / Enterococcus samt 18 stammar från olika arter.Stammarna inokulerades: - direkt på Granada agar - och efter anrikning över natten i TODD HEWITT +
antibiotikabuljong. Efter inkubering i 18-42 timmar vid 37°C under anaeroba förhållanden, observerades typiska kolonier.

RESUMÉ OG FORKLARING Granada™ Agar er et selektivt medium til screening af kliniske prøver og direkte identifikation af Streptococcus agalactiae, hos gravide kvinder og nyfødte (1, 2, 3, 4). Mediet er første gang beskrevet af dr. De La Rosa m.fl. Det er udviklet på baggrund af tidligere arbejde udført af A.K.M.S. Islam (8) og dr. De La Rosa (9, 10). S. agalactiae er ansvarlig for alvorlige infektioner i nyfødte (meningitis). Det er særligt vigtigt at opdage disse infektioner for at forebygge, behandle og overvåge dem.
PRINCIPGranada Agar er baseret på den originale formel beskrevet af dr. De La Rosa: - Granada Agar består af en næringsbase, der er en
kombination af forskellige peptoner, pyruvater og glykose.
- Methotrexaten, hesteserum og stivelsen til stede i mediet, muliggør produktionen af et pigment, der går fra orange og over i rødt, som er karakteristisk for gruppe B-streptokok-kolonier. (5)

OPBEVARINGSBETINGELSER • Pladerne opbevares ved 2-8°C i deres æske, indtil
udløbsdatoen.• Hvis pladerne ikke opbevares i æsken, kan de
opbevares i cellofanposen i 2 uger ved 2-8°C.
PRØVERPrøvemateriale består af anovaginale prøver og urinprøver fra gravide kvinder eller indtaget gastrisk væske fra nyfødte. AFLÆSNING OG FORTOLKNING • Efter inkubationen observeres bakterievæksten og
koloniernes udseende. Typiske kolonier af Streptococcus agalactiae har en farve, der går fra orange og over i rødt.
• Vækst, af andre mikroorganismer hæmmes, eller de producerede kolonier har en anden farve.

METODENS BEGRÆNSNINGER • Væksten afhænger af den enkelte individuelle
mikroorganismes behov. Det er derfor muligt, at visse stammer, som har specifikke behov (substrat, temperatur, inkubationsbetingelser etc.), ikke udvikler sig.
• I overensstemmelse med de analyserede prøver kan der medtages en ikke-selektiv blodagar.
• Nogle stammer af S.agalactiae producerer måske ikke typiske kolonier, især ikke-hæmolytiske stammer (2,3 %) (6).
• Nogle stammer, som er resistente for antibiotika i mediet, kan udvikles uden at producere typiske kolonier.
• Den orange farve kan være af varierende intensitet afhængig af stammen.
• Hvis der skal udføres en komplimentær identifikationstest med de isolerede kolonier på Granada agar, kan det ikke anbefales at bruge VITEK® GPI (ref. V 1305) og VITEK® 2 GP (ref. 21 342) reagenser.
PRÆSTATION Præstation blev vurderet ved 37°C ved hjælp af 55 stammer: 26 hæmolytiske S.agalactiae stammer, 5 ikke-hæmolytiske S.agalactiae stammer, 6 Streptococcus / Enterococcus og 18 stammer fra forskellige species. Stammerne blev inokuleret: - direkte på Granada agar - og efterfølgende beriget natten over i TODD HEWITT
+ antibiotika-bouillon. Nedenstående typiske kolonier blev observeret efter 18-42 timers inkubation ved 37°C under anaerobe forhold

Po raz pierwszy pod o ezosta o opisane przez dr De La Rosa i in. Opracowano je na podstawie pierwszej pracy A.K.M.S Islam (8) i dr De La Rosa (9, 10). S. agalactiae jest odpowiedzialny za ci kie infekcje u noworodków (zapalenie opon mózgowo-rdzeniowych). Wykrywanie go ma szczególne znaczenie w zapobieganiu, leczeniu i monitorowaniu zaka e .

Szczepy posiewano: - bezpo rednio na agar Granada - i po ca onocnym namna aniu na bulionie TODD
HEWITTA + antybiotyki. Po 18-42 godzinnej inkubacji w 37°C w warunkach beztlenowych, obserwowano wzrost typowych kolonii.
