PRESENTATION DU PROJET DE THESE
Intitulé du projet de thèse : Compréhension des mécanismes d’action d’un complexe de monoglycérides et
acides gras inhibiteurs de bactéries potentiellement pathogènes et application du complexe ou de variantes
multi-composés dans des films actifs.
Le projet s’inscrit dans le cadre des accords de coopération mis en place par le carré international
Préciser lequel :
Un accord-cadre entre l’Université de Reggio et Emilia (Unimore) et l’Université de Caen Normandie a été établi
en 2016, à l’occasion de la mise en place de la première édition de l’université d’été des apprentis en agro-
alimentaire de l’ESIX Normandie (accueil des apprentis par Unimore). Cet accord est valable 5 ans et sera
complété par de nouvelles actions dont une demande d’accueil de chercheur-invité pour le Prof. Andrea
Pulvirenti sur l’année universitaire 2019-2010.
Le projet est développé dans l’un des axes transversaux de l’établissement :

Context: Microorganisms, in particular of food origin and having a Qualified Presumption of Safety (QPS) status, are an important reservoir of ingredients either as ferments or as producers of compounds having various activity within food matrices. The ability of these microorganisms to produce volatile microbial compounds (CMVs) has been known for decades, but their biological functions and antimicrobial activities have received little attention until recently. Various studies revealed that microbial volatiles can act as infochemicals in long-distance crosskingdom communication as well as antimicrobials in competition and predation (Avalos et al, 2018), so that they play an important ecological role in the regulation of species interactions within a microbiota. Some CMVs are capable of modulating growth, virulence and biofilm formation, production of specialized metabolites (eg toxins), antibiotic resistance and spore germination in competing microorganisms. CMVs belong to various classes of

chemical compounds, including alkanes, alkenes, alcohols, esters, ketones, terpenoids, sulfur-containing compounds and inorganic compounds, and their diversity is such that there are still many CMVs to discover (O’Brien and Wright, 2011). CMVs can be unique to a single phylogenetic group or even a single species (Tyc et al, 2017), allowing their use for the selective detection of pathogens in indoor and outdoor environments and making them excellent indicators of the quality of food. Among these new compounds, some could also be used to fight against human and animal pathogens. Bacterial resistance to antibiotics is a natural phenomenon, but the spreading of multidrug resistance has been
worsened by their abuse and misuse in medicine and in farming. The biggest danger is that current antibiotics
are not able to eradicate the resistant strains. In recent years, alternative antibacterial substances have been
sought, which can help in these cases. Fatty acids and monoglycerides are known among the natural substances
for their antimicrobial properties and, important detail, bacteria do not develop resistance to them. In a previous
work at Unimore, a blend of fatty acids and their monoglycerides was created and its antimicrobial activity was
evaluated against 37 strains of Escherichia coli and 17 enterococci presenting resistance to at least two antibiotics
(Anacarso et al, 2018). Promising results were obtained with these alternative substances thus opening possible
applications to food-related Gram-positive and Gram-negative bacterial pathogens, but the mechanisms
involved in inhibition have to be elucidated.
The objectives of the project are to understand the mechanisms of action determining antimicrobial activities of
these volatile molecules and to propose and test applications relevant for the agri-food sector, and in particular
dairy and cider sectors in a transversal approach, from the cows’ udder and apples to dairy and cider
manufacturing environments. To do this:
- Starting from the UCMA microbial collection (Unicaen) the antimicrobial spectrum of the blend from
Unimore or its optimized versions will be determined by using a large collection of target microbial
species and strains relevant for the dairy and cider sectors, including Salmonella sp, Escherichia coli,
Listeria monocytogenes and Staphylococcus aureus, Zymomonas sp, Dekkera sp,
- Physiological, biochemical and molecular responses (including transcriptomic, proteomic and
metabolomics responses), cell morphology and membrane organization of representative strains will be
studied in order to describe mechanisms involved in the inhibitory activity,
- Finally, potent applications through the introduction of the mix or its optimized versions within food-
grade films will be explored in udder and stainless steel models.
In the event of satisfactory results, the perspectives of this project will be the development of antimicrobial films
to be evaluated as protective agents for teat skin in dairy cows and for apple surface, and the development of
spray to be tested for sanitation of farming/industrial equipment. Considering the components of the blend as
indicators of inhibitory activity, a screening methodology could also be developed in order to test the UCMA
microbial collection and identify potential protective strains. We want to furtherly broaden the use of natural
coatings and bioactive molecules in all the possible fields of agri-food sector.
Different formulations with bioactive products are currently studied to promote animal health and plant defense,
to reduce the abuse of antibiotics and the use of pesticides. Effective molecules have already been created during
natural evolution, we just need to find them and apply them to our human needs The present mix or its optimized
versions could be tested for their clinical application as topical treatments or, after an evaluation about the
gastric protection, as food supplements.
Scientific relations with the partner structure: this co-supervision would strengthen relations between the two
partner structures, already initiated through:
- Offers of internships at Unimore for students from Unicaen (5 students hosted in the last six years),
- Oganization of a summer university at Unimore (in 2016 and 2018) destined to students from Unicaen
(ESIX),
- Seminars conducted by Prof. A Pulvirenti at ESIX Normandie (M2 level) in 2015, 2018 and 2019,
- A request for funding to invite Prof. A Pulvirenti within Unicaen will be submitted in May 2019.
This could help to enlarge the present agreement signed by Unimore and Unicaen.

Avis motivé du directeur de thèse sur la recherche envisagée en ce qui concerne les objectifs scientifiques et technologiques poursuivis et leur place dans les orientations de recherche du laboratoire d'accueil : Les recherches menées au sein de l’équipe MALIM de l’E.A. 4651 ABTE visent à mieux connaitre le microbiote
des aliments afin d’accompagner la production d’aliments de qualité durable. Les interactions des
microorganismes entre eux, et avec leur matrice alimentaire, sont explorées pour définir des marqueurs de
qualité et/ou d’authenticité des aliments. L’équipe MALIM s’intéresse notamment à la production par les micro-
organismes de composés à activité antimicrobienne pour le contrôle des flores d’altération et/ou pathogènes au
sein des matrices d’intérêt alimentaires. Les objectifs scientifiques et technologiques de la thèse trouvent tout à
fait leur place dans les orientations de recherche du laboratoire.
Avis du directeur d’unité sous l’angle des thématiques de recherche du laboratoire et des conditions d’accueil
du candidat :
Le projet s’intègre dans l’axe de caractérisation des propriétés probiotiques des microorganismes d’intérêt
alimentaire de l’ER MALIM du projet scientifique de l’UR ABTE tel qu’évalué par l’HCERES. Il s’appuie sur le réseau
déjà constitué par le laboratoire en région avec les acteurs socio-économique du domaine agroalimentaire. Cette
thèse en co-tutelle constituera un levier de développement stratégique pour l’UR par le renforcement de sa
visibilité internationale.
Mon avis est très favorable.
Avis motivé du directeur de l’Ecole doctorale (qualité de l’encadrement et politique doctorale du laboratoire) : En co-direction avec une équipe dynamique (Pr Andrea Pulvirenti) d’Unimore (qui bénéfice d’un accord-cadre
avec Unicaen), le Pr. Desmasures propose un sujet de thèse de microbiologie, dont l’intérêt alimentaire est ancré
dans le territoire normand. Les 2 directeurs proposés ont une expérience significative et réussie de la direction
de thèse en France et en Italie. Le Pr A Pulvirenti est à cet égard directeur de l’école doctorale des Sciences agro-
alimentaires, technologies et biotechnologies d’Unimore. L’UR ABTE est très bien intégrée à l’ED nBISE et les
docteurs qui y ont été formés ont bénéficié d’un très bon encadrement. Avis très favorable.

and inhibitors of fatty acids in active films.
The Department of Life Science, University of Modena and Reggio Emilia, is also available to host the
Doctoral Candidate for future periods of study.
